How does an exosome purification method change contaminant risk?

Ultracentrifugation, filtration, size exclusion, precipitation and affinity methods do not produce the same EV mixture. Higher yield may co-recover proteins, lipoproteins, culture-medium particles and unintended vesicles; a method name is not a purity percentage.

Methods have different selectivity

Differential centrifugation, density gradients, tangential-flow filtration, size-exclusion chromatography, polymer precipitation and surface-marker capture select different physical features. A high-yield method may retain more soluble protein or lipoprotein, while a selective method can lose a target EV subgroup. MISEV2023 makes clear that saying isolated exosomes without the workflow and recovery-purity tradeoff is insufficient.

Measure purity on several axes

Consider total particles, protein, lipid, EV-enriched markers and negative markers from cells or medium together. An electron image may show attractive round structures without establishing amount or function. A comparative laboratory study found that different isolation workflows generated preparations with distinct protein loads and enzyme activity. One CD63 result cannot exclude serum lipoprotein, free cytokine or protein aggregate.

Connect method to lot and scale

When a research-tube workflow becomes thousands of commercial doses, flow rate, membrane, column, time and storage may change. Critical process parameters and acceptance ranges for yield, purity, potency and impurities need consistency across lots. The certificate must match the administered lot and final formulation. A 99% pure claim has no clinical meaning until the denominator, assay and reference material are defined.
